Ivashkiv, at the   Hospital for Special Surgery affiliated with Weill Cornell Medicine in New York City. The main focus of the lab is to     investigate cytokines, trained immunity, and the molecular, cellular and epigenetic mechanisms underlying the     pathogenesis of inflammatory and musculoskeletal diseases. The lab uses state-of-the-art molecular and genomic   approaches, preclinical models, and patient-derived samples.   Responsibilities:   The fellow will be responsible for working in conjunction with Dr. Ivashkiv to develop and carry out scientific projects related to cytokine crosstalk in inflammation and inflammatory and musculoskeletal diseases. She/he will design and perform experiments that utilize molecular, cellular and genetic techniques to answer questions about mechanisms that regulate cytokine production during inflammation. The projects will involve studying primary human macrophages from blood or synovium of healthy subjects or rheumatoid arthritis patients, and preclinical models of arthritis and inflammation, to understand signaling pathways and epigenetic mechanisms that control the expression of disease-causing genes. She/he will independently design and perform experiments that are expected to lead to first author peer-reviewed publications.   The fellow will contribute to the preparation of manuscripts and grant applications, will publish their work in high quality and impact international peer-reviewed journals, and will assist with training and supervision of technical personnel and students. She/he will maintain accurate and detailed records and databases of all laboratory work and experiments. The fellow will present at lab meetings and represent the lab at other local, national, and international forums.   The fellow will be supervised by the laboratory PI Dr. Lionel Ivashkiv. The PI will have biweekly formal meetings with the fellow to discuss the experiments, data, and plans in addition to supervision and mentoring during lab meetings and informal meetings to discuss data or troubleshoot problems.

Working with colleagues who love what they do and are deeply committed to our Mission, you too can be part of our transformation across the enterprise.   Postdoctoral Fellow Bostrom Lab Full-Time   Position Overview:   Postdoctoral Fellow position is available in the Arthroplasty Research Lab of Dr. Mathias Bostrom at Hospital for Special Surgery Research Institute (HSSRI), to understand how stem cell mediate bone regeneration and orchestrate a composite tissue regeneration program.   This project will be mainly supported by a newly funded NIH R01 with Dr. Matthew Greenblatt of Weill Cornell Medicine and Dr. Xu Yang of the Arthroplasty Research Lab at HSSRI as Co-PIs. We will develop key initial elements of a new approach for the regeneration of amputated limbs, by transducing a specific population of autologous skeletal stem cells to induce a blastema-like state, and then implanting them into the amputation site to initiate regeneration of the missing limb.   The candidate may also participate in other projects to invest the cellular and molecular mechanism of orthopaedic implant osseointegration, including an on-going project funded by Department of Defense, with Dr. Xu Yang as the PI.   Candidate having a recent Ph.D. degree in Developmental Biology, Cell and Molecular Biology, or Neuroscience will be desirable. Writing skill and expertise in mouse genetics, light and confocal microscopy, and gold-stand techniques of cell and molecular biology are required. The candidate is expected to have one or more high quality first author publications on cell biology or mouse models of disease or both. The candidate should be highly motivated and productive and interested in a career in research. Expertise in mouse models for wound healing and/or in working with skeletal stem/progenitor lineage cells would be a plus. The postdoctoral fellow will report directly to Dr. Xu Yang to discuss the experimental plan and research direction in the area of interest and attend the weekly lab meeting with Dr. Bostrom and Dr. Greenblatt.   Major duties and responsibilities - Design and execute experimental procedures/protocols in research involving cell and molecular biology techniques, tissue culture, histology and immunological assay, polarizing and fluorescence microscopy, and animal model system including mouse breeding and genotyping. - Collecting, analyzing and reporting on experimental data to the Principal Investigator on a timely manner. - Maintain accurate and detailed records and databases of all laboratory work and experiments. - Apply standard scientific protocols to experiments, research and improve current methods, and develop, set-up, and verify new procedures in the - Standardize and establish new published procedures; evaluate innovative techniques. Develop new assays and new protocols for research. - Maintain optimal protocol and assay accuracy by conducting regular quality and reproducibility checks. - Timely compile, present and discuss relevant insights about experimental results with Principal Investigator. - Maintain and monitor proper use of laboratory equipment. - Troubleshoot and correct erroneous results or problems with laboratory equipment. - Draft, write, and edit scientific papers, journal articles, abstracts and progress reports related to the work. - Maintain an integral role in the laboratory's meetings, departmental seminars, presentations, and publications. - Organization, and recording of research material inventory, and placing order for research reagent in a timely manner - Completion of all trainings related to work in a timely manner, including C14, animal usage, handling hazardous material, laboratory safety. - Participate in establishing job requirements and goals; perform duties at the desired level of competency.

Working with colleagues who love what they do and are deeply committed to our Mission, you too can be part of our transformation across the enterprise.   Postdoctoral Fellow Bostrom Lab Full-Time   Position Overview:   Postdoctoral Fellow position is available in the Arthroplasty Research Lab of Dr. Mathias Bostrom at Hospital for Special Surgery Research Institute (HSSRI), to study the role of immune cells in successful and failed osseointegration.   Using a set of mouse implant osseointegration models developed by Dr. Xu Yang of the Arthroplasty Research Lab, we have recently demonstrated that LepR+ cells are the origin of peri-implant fibroblasts. The paper reporting this finding has been accepted by Nature Biomedical Engineering. We will now use these models to study the interactions between immune cells and skeletal stem/progenitor cells after implantation. A special focus will be put on the mechanism and consequence of the formation of fibrosis tissue around implant during failed osseointegration. Human macrophages or skeletal stem/progenitors from bone marrow or periprosthetic tissue of patients receiving primary or revision joint replacement surgeries will also be used to direct the design of animal models or confirm the findings from animal models.   Dr. Xu Yang is also collaborating with Dr. Lionel Ivashkiv, a bone immunologist, at HSSRI on a NIH R01 grant to investigate the inflammatory and bone marrow responses that drive osteoclastogenesis, and the feedback mechanisms that suppress the adaptive immune response to periprosthetic joint infection.   The position requires a Ph.D. in Immunology or Molecular Biology, who has direct experience working with immune cells using advanced cellular, molecular, and genomic techniques. This position combines in vivo research with molecular and cellular biology. For in vivo work, the candidate will be capable of assisting animal surgeries and have experience with isolating proteins and nucleic acids from mouse bone and clinical samples from patients receiving joint replacements. For cellular and molecular studies, the candidate will also have experience with flowcytometry, immunofluorescence, cell culture, RT-PCR, and RNA-seq. The candidate is expected to have one or more high quality first author publications on immunology or mouse models of inflammation disease or both. The candidate should be highly motivated and productive and interested in a career in research. The successful candidate will work on existing animal models of bone healing and ectopic mineralization, for the purposes of lineage tracing, functional manipulation and in-depth cellular characterization. Responsibilities include: - Develops and maintains research databases with guidance from Principal Investigator - Performs data collection and data entry for studies as requested.  Schedules and conducts subject interviews, follow-up visits and phone calls to obtain data as per protocol.  Ensures accuracy when transcribing data from data collection sheets, medical records, or electronic institutional databases or other source documents into databases. - Performs or helps facilitate informed consent procedures following all regulatory guidelines.  - Participates in protocol development in collaboration with Principal Investigator and co-investigators. Conducts literature searches; actively participates in development meetings; reviews and edits protocol document; prepares study initiation documents. - Prepares drafts of protocols and manuscripts as requested by the Principal Investigator. - Performs IRB submissions for initial approvals, re-approvals, amendments, etc.  Prepares, submits and files IRB and all regulatory documents, ie., data collection sheets, protocol amendments, protocol deviations, adverse event forms, etc. - Prepares information required for reports to research administration as needed. - Ensures that all research related regulatory, institutional, and departmental compliance requirements are met as appropriate. - Conducts statistical data analysis and prepares statistical graphs and tables in preparation for research presentations, abstract and manuscript preparation or for other publications as requested by the Principal Investigator.
